ORIC Pharmaceuticals

Clinical-stage oncology company targeting resistance

Overview

ORIC Pharmaceuticals is a clinical-stage cancer company focused on overcoming how cancers resist treatment. Its therapies aim to restore patients’ response to existing treatments by targeting resistance mechanisms. Lead programs include rinzimetostat (ORIC-944), an allosteric inhibitor of the PRC2 complex designed for prostate cancer, and enozertinib (ORIC-114), a brain-penetrant inhibitor that targets EGFR exon 20 and HER2 exon 20 mutations in non-small cell lung cancer. Early work continues on other resistance mechanisms across solid tumors. Unlike some competitors, ORIC concentrates specifically on breaking or bypassing resistance to current therapies, based on research from Memorial Sloan Kettering Cancer Center and brought to Nasdaq-listed status in 2020. Its goal is to improve how well existing cancer treatments work for patients by keeping cancer from outsmarting them.

ORIC(R) Pharmaceuticals announces agreement with Cancer Research UK to advance a potential first-in-class cip2a-targeted therapy into clinical development.

ORIC(R) Pharmaceuticals announces agreement with Cancer Research UK to advance a potential first-in-class cip2a-targeted therapy into clinical development. ORIC-259 is a potential first-in-class small molecule mRNA degrader targeting CIP2A, a previously undruggable target Cancer Research UK to fully fund, sponsor, design, and execute CTA-enabling studies and Phase 1/2 clinical development ORIC retains exclusive option to reacquire rights to ORIC-259 following Phase 1/2 data SOUTH SAN FRANCISCO, Calif. and SAN DIEGO, Oct. 05, 2026 (GLOBE NEWSWIRE) - ORIC Pharmaceuticals, Inc. (Nasdaq: ORIC), a clinical stage oncology company focused on developing and commercializing treatments that address mechanisms of therapeutic resistance, today announced an agreement with Cancer Research UK, one of the world's largest funders of cancer research, to advance a potential first-in-class CIP2A-targeted therapy into clinical development. Under the agreement, Cancer Research UK's Centre for Drug Development (CDD) will fully fund, sponsor, design, and execute CTA-enabling studies and early clinical development of ORIC-259 for patients with cancer. ORIC retains an option to reacquire rights to ORIC-259 following Phase 1/2 data, with Cancer Research UK eligible to receive downstream milestone payments and royalties. Cancerous Inhibitor of PP2A (CIP2A) is a protein that contributes to the development and progression of many cancer types due to its role in protein phosphorylation, mitosis, and DNA damage response. ORIC-259 is a potent, selective, orally bioavailable small molecule degrader of CIP2A mRNA. This potential first-in-class mRNA degrader is designed to modify mRNA splicing to incorporate a premature stop codon, leading to mRNA degradation and loss of CIP2A protein, which is critical for cell proliferation and survival of cancers with homologous recombination deficiency (HRD). Promising preclinical data highlight robust antitumor activity in PARP inhibitor sensitive and resistant settings, indicating broad potential for patients with HRD cancers, and supports the path to clinical development for ORIC-259. "We are excited to partner with Cancer Research UK to advance ORIC-259, a potential first-in-class CIP2A-targeted therapy, into the clinic," said Lori Friedman, PhD, chief scientific officer. "Cancer Research UK's proven expertise and capabilities in developing innovative cancer therapies make it an ideal partner for the program, while this structure allows us to remain disciplined in allocating our internal resources to our late-stage clinical pipeline." "ORIC-259 represents an innovative approach to targeting CIP2A, a protein with significant potential in cancer but one that has proven difficult to drug," said Lars Erwig, Director of the CDD. "We share ORIC's mission to overcome treatment resistance in cancer, and we look forward to applying our expertise to explore new treatment options for patients." About ORIC Pharmaceuticals, Inc. ORIC Pharmaceuticals is a clinical stage biopharmaceutical company dedicated to improving patients' lives by Overcoming Resistance In Cancer. ORIC's clinical stage product candidates include (1) rinzimetostat, an allosteric inhibitor of the polycomb repressive complex 2 (PRC2) via the EED subunit, being developed for prostate cancer, and (2) enozertinib, a brain-penetrant inhibitor targeting EGFR exon 20 insertion and EGFR atypical mutations, being developed for NSCLC. ORIC Pharmaceuticals narrows loss per share despite $77M net loss as $12M ESOP filing signals ongoing dilution risk

ORIC Pharmaceuticals reported a wider net loss of $41.49 million for Q2 2026 and $77.26 million for the first half, though basic loss per share from continuing operations narrowed year-over-year. Days earlier, the biotech filed a $12.41 million shelf registration for 1.1 million common shares under an ESOP-related offering. The company has no revenue and continues burning cash whilst advancing its oncology pipeline, including rinzimetostat in the Phase 3 Himalayas-1 trial and earlier-stage programmes like enozertinib. The narrower per-share loss reflects share count dynamics rather than operational improvement. Community fair value estimates range widely from $4.34 to $43.40 per share, highlighting divergent views on ORIC's prospects given its zero revenue and reliance on future trial success. Recent price strength suggests the market is focusing on pipeline milestones rather than near-term accounting metrics.

ORIC Pharmaceuticals has $282M cash and 2.5-year runway despite $111M annual burn

ORIC Pharmaceuticals has $282 million in cash with no debt as of March 2026, providing approximately 2.5 years of cash runway based on its annual cash burn of $111 million. The biotech company's cash burn decreased by 4.4% over the past year. As a pre-revenue, early-stage company, ORIC Pharmaceuticals is still developing its business. The reduced cash burn suggests management is maintaining steady business development whilst slightly decreasing expenditure. Whilst the current cash runway appears prudent, shareholders may need to consider how easily the company could raise additional capital through issuing new shares or taking on debt if required for future growth.

ORIC Pharmaceuticals shares jump 19% after promising rinzimetostat prostate cancer data

ORIC Pharmaceuticals has released promising Phase 1b data for rinzimetostat plus darolutamide in metastatic castration resistant prostate cancer, selecting a 400mg daily dose for a planned global Phase 3 trial. The company's shares rose 18.93% in one day to $8.89, though this followed a 27.07% seven-day decline. Despite the volatility, ORIC's one-year total shareholder return stands at 93.68%. The stock trades well below the average analyst price target of $21, with a DCF model suggesting a fair value of $43.31 per share. However, the clinical-stage biotech currently has no revenue and reported a net loss of $129.468m. The company's price-to-book ratio of 2.3x appears reasonable compared to peers at 4.7x, though slightly above the US biotechs average of 2.2x.
