Responsibilities

Establish the upstream process of cell culture and AAV vector production in single-use or multi-use bioreactors. Optimize the downstream process of large-scale vector harvest, purification and formulation
Work closely with other members of the Viral Delivery Team to establish PD-related late-stage AAV analytics assays
Accurately and timely record, document, analyze and communicate laboratory data to senior staff and team members
Conduct all activities in a safe and efficient manner, and adheres to site environmental health and safety (EHS) requirements
Supervise research associates as needed

Desired Qualifications

Experience in AAV vector PD and regulatory policy for late-stage manufacturing
Experience in depth filtration (DF) of AAV crude lysates
Experience in ion-exchange (IEX) chromatography
Experience in AAV analytics assays for quantification of AAV titers, empty/full particle ratio, host cell impurities, and process-related impurities
Familiar with AAV vector biology and new vector design
Postdoc experience is preferred for PhD candidates
Experience with Benchling, Microsoft Office Suite and other data analysis software (SnapGene, GraphPad Prism, etc.)
Strong organizational and record-keeping skills
Excellent interpersonal, verbal and written communication skills
Highly motivated with strong work ethic
People management experience

OPPORTUNITY

Mammoth is currently seeking a talented Senior Scientist I with experience in AAV process development (PD) to join our Viral Delivery Team. The successful candidate will work closely and collaboratively with colleagues to design and perform experiments on large-scale AAV vector production and purification to support AAV delivery of Cas nucleases for human therapeutics. The position requires adaptability, excellent technical and analytical skills, strong organizational and communication skills, as well as the ability to work independently in a fast-paced team-oriented and dynamic environment. May supervise research associates.

TEAM

Our small-size proteins, such as Cas14 and CasPhi, are poised to transform disease treatment enabling novel editing approaches and delivery options that are difficult to achieve or not even possible with previously described CRISPR-Cas systems.
