Requirements
  • Candidates must have extensive hands-on experience generating transgenic mouse models using CRISPR/Cas9 gene editing, including knockouts, conditional knockouts, single-point mutations, insertions, and gene fusions, and simultaneous multi-gene modifications
  • Proven experience generating transgenic mice by injecting plasmid or bacterial artificial chromosome DNA constructs into inbred and outbred mouse strains
  • Experience with knockout, conditional knockout, and knock-in generation using traditional embryonic stem cell targeting, including C57BL/6 embryonic stem cells
  • Proficiency in embryology techniques such as embryo microinjection, ES cell microinjection, rederivation, embryo and sperm cryopreservation, in vitro fertilization, tail biopsy, and genotyping
  • Demonstrated ability to manage and maintain transgenic mouse colonies, including breeding program design, genotyping, phenotyping, pedigree tracking, scaling colonies, and minimizing genetic drift
  • Strong recordkeeping, data analysis, and reporting skills; ability to develop standard operating procedures and ensure compliance with institutional, national, and international guidelines for animal research (IACUC and AAALAC) and workplace safety
  • Leadership or supervisory experience overseeing project teams and coordinating resources, budgeting, and quality control
Responsibilities
  • Develop and execute strategies for generating transgenic mouse models to meet project-specific research goals
  • Employ CRISPR/Cas9, pronuclear injection, homologous recombination, and other genome-editing tools to design custom mouse lines in coordination with director and research teams
  • Model Creation: perform embryo microinjections and other micromanipulation techniques to introduce genetic modifications; optimize protocols for high efficiency; troubleshoot challenges
  • Colony Management: design and oversee breeding programs to establish, expand, and maintain transgenic mouse colonies; ensure accurate genotyping, phenotyping, and pedigree tracking; implement strategies for colony scaling
  • Model Characterization: characterize transgenic mice for gene integration, expression, and phenotypic outcomes; use bioinformatics tools to analyze data; provide interpretation to stakeholders
  • Recordkeeping, Reporting and Compliance: maintain records; generate detailed reports; develop SOPs; ensure compliance with guidelines (IACUC, AAALAC) and welfare
  • Scientific support, Collaboration and Innovation: conduct/support research; advise/train personnel; foster collaborations; provide training; stay informed on emerging technologies; develop new services; participate in conferences
  • Resource and Facility Management: oversee maintenance of equipment, facilities, reagents; coordinate procurement and budgeting; manage activities with cost effectiveness; implement quality control
  • Others: adhere to policies; safeguard assets; perform other duties; note management reserves right to revise
