The Role:
Generate:Biomedicines is seeking an exceptional and highly motivated scientist with deep expertise in mass spectrometry and a proven track record of analytical characterization of diverse biological macromolecules, including monoclonal antibodies, bi/multi-specifics, Fc-fusion proteins, peptides and enzymes with a strong desire and potential to learn and grow in a fast paced and innovative working environment. The responsibilities encompass, and are not limited to, performing high throughput (HTP) and detailed analytical characterization of early drug candidates for verification of molecular weights, protein sequence, and the identification and quantitation of post-translational modifications including glycosylation by mass spectrometry. The candidate must be well versed in a range of mass spectrometry based methods and techniques including intact protein analysis, peptide mapping, and structural mass spectrometry. They will be responsible for performing fit-for-purpose method development and contribute to the development and growth of new LC-MS based platforms to support the characterization and development of novel biotherapeutic molecules. They must keep current on advances and developments in LC-MS instrumentation and applications and contribute to the growth of the Protein Analytics Mass Spectrometry Group’s capabilities. They will contribute to HTP LC-MS method development, execution, and data analysis in support of our machine learning platform. They will also contribute to platform and project advancement through interfacing with project teams and managing deliverables related to the Protein Analytics team’s workflows.
Here is how you will contribute:
- Develop and execute LC-MS methods for the analytical and structural characterization of proteins.
- Perform routine HTP LC-MS data collection, analysis, and interpretation and be accountable for communicating results to stakeholders.
- Establish and lead cross functional collaborations.
- Drive the exploration and adoption of new technologies and platforms.
- Author internal operation protocols and/or external publications.
- Potential for project or team leadership opportunities.
The Ideal Candidate will have:
PhD in relevant field with 0-2 years, or MS/BS with 8+ years experience in Biotech/Pharm
Extensive experience with:
- Operation and maintenance of the latest mass spectrometry instrumentation from multiple vendors (Thermo Exploris or Q-Exactive series, Thermo Fusion Lumos or Eclipse, Waters Synapt or Xevo, Waters BioAccord, Agilent 6545XT, etc.)
- Mass spectrometry method development and troubleshooting
- Protein and peptide separation by reverse phase chromatography by UPLC, UHPLC, nano-, micro-, and capillary-flow LC
- Reducing and non-reducing peptide mapping
- Protein characterization by intact mass spectrometry
- Expertise in mass spectrometry data analysis tools (Agilent BioConfirm, Protein Metrics Byos and Byonic, BioPharma Finder, Proteome Discoverer, Skyline, MaxQuant, Unidec, and PEAKS)
- Excellent attention to detail and documentation skills
- Demonstrating outstanding communication skills, transparency and respect for others
Preferred Qualifications:
- Educational background in protein chemistry/biochemistry/biophysics is strongly preferred.
- Working experience with:
- Native MS
- Structural mass spectrometry by HDX, crosslinking, FPOP, or HRPF
- LC-MS based proteomics approaches including; quantitative proteomics, targeted proteomics, immunopeptidomics, and spatial/structural proteomics.
- Glycan typing/quantifying by HLIC
- Multiplex MS via isotopically-labeled tags (e.g. TMT)
- Working knowledge in ion-mobility MS
- Experience in, or insight into, bioanalytical mass spectrometry for PK/PD is a plus.
- Knowledge of R/Python (or similar) languages is a plus
