Requirements
  • A PhD in Chemistry, Pharmaceutical Chemistry, or a related discipline, plus 1–2 years of relevant work; or an MS with five years of focused lipid design experience for liposomal or lipid nanoparticle applications.
  • Strong skill in designing and synthesizing new lipids or other delivery chemistries through hands-on execution.
  • Experience with structure–activity relationship-driven design, synthesis, purification, and characterization of ionizable lipids and polyethylene glycol-lipid conjugates for targeted lipid nanoparticle applications, using ChemDraw, SciFinder, Reaxys, and artificial intelligence-based structure–activity relationship prediction tools.
  • Working knowledge of conjugation chemistries for attaching peptides, antibodies, nanobodies, and small-molecule ligands, including linker design and selection of suitable biorthogonal or site-specific handles.
  • Practical purification skills using normal-phase and reverse-phase chromatography and preparative high-performance liquid chromatography.
  • Analytical characterization experience with liquid chromatography charged aerosol detection, liquid chromatography ultraviolet detection, liquid chromatography tandem mass spectrometry, and nuclear magnetic resonance spectroscopy, including 1H, 13C, and 31P as needed, with the ability to deconvolute data and produce well-characterized lipids.
Responsibilities
  • Design new ionizable lipids and lipid conjugates for antibodies, peptides, and other targeting moieties across multiple structural families to advance the platform beyond current-generation materials.
  • Devise and execute practical synthetic routes to priority targets to support pipeline needs for targeting lipids.
  • Plan synthesis and purification for these modalities with scale and final purity in mind, delivering against agreed project timelines.
  • Build ionizable and targeting lipids for the targeted lipid nanoparticle platform using a structure–activity relationship-based approach to guide the next design cycle.
  • Develop conjugation chemistry linking peptides, antibodies, nanobodies, and small-molecule ligands to lipid anchors to generate building blocks for cell- and tissue-selective delivery.
  • Translate in vitro and in vivo discovery data into concrete structural hypotheses and revised molecules.
  • Present synthetic strategy, experimental results, and structure–activity relationship conclusions, and author protocols and technical reports.
Desired Qualifications
  • Familiarity with lipid degradation pathways and impurity work, including N-oxides, hydrolysis products, and lipid–nucleic acid adducts, and their effects on potency and stability.
  • Solid-phase peptide synthesis or antibody/nanobody modification experience.
  • Exposure to scale-up, chemistry, manufacturing, and controls, or technology transfer to a contract development and manufacturing organization.
  • Experience with data visualization and statistical analysis using R, Python, or JMP.
