Kura Oncology has launched Caspian Therapeutics with $50 million in financing to develop menin inhibitors for diabetes treatment. The financing was led by BVF Partners, with participation from T1D Fund, Invus, Montanova, Eli Lilly and Company, and Kura Oncology itself. Caspian will advance KO-7246, a next-generation menin inhibitor designed for chronic metabolic diseases. Preclinical studies show the compound can selectively expand pancreatic β cells, increase insulin production, and improve blood sugar control in Type 1 and Type 2 diabetes models. Kura Oncology retains approximately 50% ownership of Caspian on a fully diluted basis whilst maintaining its focus on oncology. The financing will fund KO-7246 through IND-enabling development and initial clinical proof-of-concept trials in diabetes patients. Robert Spencer has been appointed President and Chief Operating Officer of Caspian.
Eli Lilly's OLUMIANT (baricitinib) generated $957.4 million in worldwide revenue in 2024, up from $922.6 million in 2023 and $830.5 million in 2022, according to a new market analysis from ResearchAndMarkets.com. The oral JAK inhibitor is approved for alopecia areata, atopic dermatitis, rheumatoid arthritis and COVID-19 across seven major markets: the United States, Germany, France, Italy, Spain, the United Kingdom and Japan. OLUMIANT became the first FDA-approved systemic treatment for severe alopecia areata in June 2022. In pivotal Phase III trials, approximately 35 to 40 per cent of patients on the 4mg dose achieved at least 80 per cent scalp hair coverage. Growth in alopecia areata has offset pressure in mature rheumatoid arthritis markets. The report forecasts OLUMIANT sales through 2034.
Cloverleaf Bio, an RNA therapeutics company developing engineered transfer RNA-based cancer treatments, has closed a $33 million seed financing round. The oversubscribed round was led by 4BIO Capital, with participation from AbbVie Ventures, Eli Lilly and Company, Boehringer Ingelheim Venture Fund, Draper Associates, Mission BioCapital, and American Cancer Society BrightEdge. The New Haven-based company will use the funds to advance two lead programmes: an inhibitory tRNA asset targeting hepatocellular carcinoma through early clinical testing and an antibody-tRNA conjugate asset for colorectal cancer through preclinical development. Cloverleaf's platform targets RNA-modifying enzymes that cancer cells depend on during proliferation. In preclinical studies, the lead compound CLB-001 demonstrated greater potency and selectivity for cancerous cells whilst showing no signs of systemic toxicity. The company is a Yale University spinout, founded by Austin Draycott, Cole Lewis, and Wendy Gilbert.

The drop cut the stock's year-to-date gain to around 14%. The trial in question is the Lp(a)HORIZON study, which enrolled more than 8,000 patients with elevated levels of lipoprotein(a), a cholesterol-carrying particle in the blood that is linked to heart disease. Pelacarsen did succeed in lowering Lp(a) levels. But lower blood levels did not translate into fewer cardiovascular events, like heart attacks, strokes, or death. That gap between a biomarker moving and actual patient outcomes is a tough result for any drug to recover from. Chief Medical Officer Shreeram Aradhye acknowledged the disappointment, saying the results were "not what we hoped for," but added the data still advances understanding of the relationship between Lp(a) lowering and cardiovascular risk. Questions for rival drugs too. The failure does not only affect Novartis. Amgen and Eli Lilly are both developing their own drugs that target Lp(a), though through a different biological pathway. The pelacarsen outcome raises questions about whether lowering Lp(a) alone is enough to protect the heart. No drug specifically targeting lipoprotein(a) has been approved yet, despite elevated Lp(a) affecting roughly one in five people worldwide. Pelacarsen was licensed by Novartis from Ionis Pharmaceuticals. It is an antisense oligonucleotide, meaning it targets the production of Lp(a) at its source. More pressure on the pipeline. Analysts at Jefferies noted that the broader success of existing cardiovascular therapies, combined with the rise of GLP-1 obesity drugs, is raising the bar for new heart treatments to prove their value in outcome trials. The trial failure also comes less than a week after Novartis paused eight trials of a separate experimental cell therapy for autoimmune and neurological disorders following the deaths of three patients. Jefferies said recent positive data from a multiple sclerosis trial should help soften the blow for investors. But the bank also noted that Novartis now faces pressure to deliver positive results from a drug it acquired through its $12 billion Avidity Biosciences deal to justify that price tag.
