About the company

Revolution Medicines develops targeted therapies for cancers caused by RAS mutations, which are found in about 30% of human cancers. The company’s products are small molecules called RAS(ON) inhibitors that work by blocking the active, cancer-promoting form of the RAS protein to stop tumor growth. Unlike older treatments that could only target a few specific mutations, this platform creates inhibitors that can address a much wider range of RAS-driven cancers. The company's goal is to advance these candidates through clinical trials to provide effective treatment options for patients with previously "undruggable" forms of cancer.

What believers are saying

  • FDA approved RASONQUE on August 26, 2026, six months ahead of deadline.
  • RASolute 302 cut death risk 60% and doubled median overall survival to 13.2 months.
  • Royalty Pharma's June 2025 $2 billion package funds commercialization and global expansion through 2027.

What critics are saying

  • RASONQUE costs $39,800 monthly, inviting Medicare and commercial payer restrictions in 2026.
  • Revenue still depends overwhelmingly on one launch; any RASONQUE setback crushes the company.
  • AstraZeneca, Amgen, and Mirati pressure RAS programs; resistance or safety issues can erase the franchise.

What makes Revolution Medicines unique

  • RASONQUE became the first approved broad RAS(ON) inhibitor on August 26, 2026.
  • Revolution Medicines avoids companion diagnostics and treats metastatic PDAC with or without RAS mutations.
  • Its pipeline spans daraxonrasib, elironrasib, and zoldonrasib across pancreatic, lung, and colorectal cancers.

Novocure raises 2026 revenue guidance to $736M–$743M as Daraxonrasib launch slows Optune Pax

Novocure's chief executive officer Frank Leonard said the launch of Revolution Medicines' drug Daraxonrasib slowed adoption of its Optune Pax device. He said it has "significantly" limited the company's access to medical practices. Active US Optune Pax patients fell to 266 at the end of the third quarter from 285 in the second. Wedbush, which rates Novocure "Neutral," sees disruption at practices rather than "direct displacement." Third-quarter revenue rose 16% to $193.4 million, beating estimates of $182.2 million. Novocure raised its 2026 revenue guidance to $736 million to $743 million from $710 million to $725 million. Shares fell about 4% after hitting six-month lows earlier in the session.

Druckenmiller boosted Revolution Medicines stake before FDA nod; is the $40B biotech still a buy?

Revolution Medicines (NASDAQ: RVMD) has become one of this year's hottest biotech stocks after the US approved its pancreatic cancer drug, Rasonque, in August. Before the approval, Duquesne Family Office, led by billionaire Stanley Druckenmiller, raised its stake in the company by 26.5% in the second quarter. In a Phase 3 study, Rasonque achieved median overall survival of 13.2 months versus 6.7 months for chemotherapy. The drug costs about $478,000 per year at list price. Assuming a 30% net discount, the US market for second-line patients is nearly a $3 billion annual opportunity. The company's market cap is about $40 billion. The author says the stock looks more attractive than months ago, but would consider buying if the market cap falls by another $5 billion.

ESMO 2026: Rasonque nearly doubles pancreatic cancer survival to 13.2 months as market heads to $10.2B

Revolution Medicines' oral drug Rasonque (daraxonrasib) nearly doubled overall survival in pancreatic cancer, according to Phase III RASolute 302 data. Median survival was 13.2 months, against 6.7 months with standard care. The US Food and Drug Administration approved the drug in August 2026. Further findings at the 2026 European Society for Medical Oncology Congress showed Rasonque beat placebo across mutation subgroups. In the most common, RAS G12D, the overall response rate was 32% versus 11%. Tango Therapeutics reported an 82% response rate for vopimetostat combined with Rasonque, and plans a Phase III trial in the first half of 2027. GlobalData valued the pancreatic cancer market in the eight major markets at $2.2bn in 2024 and expects $10.2bn by 2034.

ESMO26: Tango sees 82% ORR with combo in pancreatic cancer.

ESMO26: Tango sees 82% ORR with combo in pancreatic cancer. At this year's ASCO congress, Revolution Medicines' Rasonque caused a stir in pancreatic cancer. Now, a study combining the drug with Tango Therapeutics' vopimetostat has delivered another positive result at ESMO. The phase 1/2 study of PRMT5 inhibitor vopimetostat with Rasonque (daraxonrasib), a RAS(on) inhibitor, revealed an impressive 82% objective response rate (ORR) when used as a second- or third-line therapy in 22 evaluable patients with MTAP-deleted and RAS-mutant metastatic pancreatic ductal adenocarcinoma (PDAC). At nine months, 88% of patients who had been treated with the dual therapy were still alive, without progression of their cancer, and the median progression-free survival (PFS) had not been reached, pointing to a durable response. A small analysis in five evaluable patients with non-small cell lung cancer (NSCLC) revealed an 80% ORR and a 100% disease control rate. Meanwhile, the combination was "generally well-tolerated" in 30 patients with PDAC and NSCLC who had received the lower dose level (200 mg once-daily) in the ongoing study, although Tango said the higher dose (250 mg) "was not well-tolerated." That safety profile may have spooked some investors, with shares in Tango slipping around 9% at the time of writing, but could equally signal profit-taking on earlier gains in the stock. The RASolute 302 trial of Rasonque in PDAC prompted a standing ovation at ASCO, recognising the desperate need for new therapies for this aggressive cancer, and the new data raises the hope that combination therapy could move the efficacy needle even further. Armed with the new data, Boston, Massachusetts-based Tango has started preparations for a pivotal phase 3 trial of vopimetostat 200 mg plus Rasonque as a front-line treatment for MTAP-deleted pancreatic cancer, hoping to get that underway in the first half of 2027. It will also start discussions with regulators about the potential for approval as a second-line therapy based on the phase 1/2 trial. Mutations or deletions in the MTAP gene are seen in between 18% and 40% of pancreatic cancers, leading to the accumulation of a metabolite (MTA) that binds to and suppresses the PRMT5 enzyme. That suppression weakens the cells, and adding vopimetostat can drive levels of the enzyme below the threshold for survival, while sparing healthy cells that retain normal PRMT5 activity. The hope is that combining PRMT5 inhibition and Ras(on) blockade can boost efficacy whilst also preventing the emergence of tumour resistance, which has been observed with Rasonque monotherapy. "In this updated analysis, the large majority of patients remained progression-free at nine months despite being heavily pretreated, with more than half receiving the combination as third-line therapy," said lead investigator Alexander Spira, of Virginia Cancer Specialists Research Institute. "Achieving this level of durability with an oral, chemotherapy-free regimen is particularly meaningful for patients and highlights the potential of a vopimetostat combination to redefine the standard of care in a disease that has been notoriously difficult to treat." 23 October, 2026
