About the job

Requirements
  • A Bachelor of Science in Chemistry, Biochemistry, Chemical Biology, or a related discipline with at least 2 years of relevant laboratory experience, or a Master of Science in a related discipline with 0–2+ years of experience.
  • Hands-on experience with solid-phase oligonucleotide synthesis on an automated synthesizer, including cleavage, deprotection, and workup.
  • Demonstrated experience with preparative high-performance liquid chromatography purification and interpretation of analytical high-performance liquid chromatography or liquid chromatography–mass spectrometry data.
  • Practical experience with at least one bioconjugation chemistry, such as N-hydroxysuccinimide ester, maleimide–thiol, or click chemistry, applied to oligonucleotides, peptides, or proteins.
  • Strong general wet-chemistry experience, including accurate weighing, solution preparation, air- and moisture-sensitive handling, and anhydrous techniques.
  • Meticulous documentation habits and comfort working in an electronic lab notebook.
  • Ability to manage multiple parallel experiments and deliver against timelines in a fast-paced environment.
Responsibilities
  • Execute phosphoramidite-based solid-phase synthesis of chemically modified RNA and DNA on automated synthesizers at micromole to millimole scales.
  • Incorporate chemically modified building blocks, including 2′-OMe and 2′-F, phosphorothioate and other backbone modifications, and 5′/3′ functional handles for downstream conjugation.
  • Perform cleavage from support, base and 2′-silyl deprotection, desalting, and lyophilization using established protocols.
  • Prepare amidite and reagent solutions, maintain synthesizer uptime through routine maintenance and troubleshooting, and manage consumable and raw material inventory.
  • Purify crude oligonucleotides by reverse-phase and ion-exchange high-performance liquid chromatography; pool, desalt, concentrate, and formulate final material to target concentration and buffer.
  • Assess identity and purity by liquid chromatography–mass spectrometry, ultra-performance liquid chromatography, and ultraviolet quantification; interpret chromatograms and deconvoluted mass data.
  • Prepare small molecule–oligonucleotide conjugates, such as GalNAc, lipid, and peptide conjugates, and support preparation of antibody–oligonucleotide conjugates using standard bioconjugation chemistries.
  • Purify conjugates by chromatography and filtration, and assess purity, conjugation efficiency, and drug-to-antibody ratio.
  • Run small-scale screens of reaction conditions and summarize results for the team.
  • Maintain complete, contemporaneous electronic lab notebook records and accurate sample registration and inventory in compound and reagent management systems.
  • Analyze and present data clearly at team and project meetings.
  • Work in accordance with environmental, health, and safety requirements, including safe handling of organic solvents and reactive reagents, proper waste disposal, and correct use of fume hoods and personal protective equipment.
Desired Qualifications
  • Experience synthesizing chemically modified small interfering RNA, antisense oligonucleotides, or guide RNA with phosphorothioate and 2′-modified nucleotides.
  • Experience preparing GalNAc, lipid, or peptide conjugates for targeted nucleic acid delivery.
  • Experience with antibody handling and antibody–drug or antibody–oligonucleotide conjugate preparation and characterization, including drug-to-antibody ratio determination.
  • Familiarity with protein purification platforms such as fast protein liquid chromatography, size-exclusion chromatography, hydrophobic interaction chromatography, and tangential flow filtration.
  • Experience with liquid chromatography–mass spectrometry deconvolution software and chromatography data systems such as Empower, Chromeleon, or Unicorn.
  • Exposure to design of experiments or structured screening approaches for reaction optimization.
  • Experience supporting material transfer to external vendors or contract development and manufacturing organizations.

About the company

CRISPR Therapeutics develops gene-editing medicines using CRISPR/Cas9 to treat serious diseases. It edits genes in human cells, either ex vivo or in vivo, to correct disease-causing mutations, focusing on sickle cell disease, cancer, and other genetically defined conditions. The company supports its pipeline through partnerships and licensing in addition to internal R&D and clinical work, rather than relying solely on in-house development. Its goal is to deliver durable, disease-modifying therapies that meet unmet medical needs for patients worldwide through a global biopharmaceutical strategy.

What believers are saying

  • FDA approved CASGEVY for children age 2+, expanding eligibility by roughly 5,500 patients.
  • Germany reimbursement on May 6, 2026 broadens access and supports European uptake.
  • Phase 1 starts for CTX340 and CTX460 create multiple new catalysts in 2026.

What critics are saying

  • CASGEVY revenue was only $76 million in Q2 2026, while losses hit $91.2 million.
  • Broad won another March 2026 PTAB ruling; CRISPR's CRISPR-Cas9 foundation remains legally fragile.
  • If slow patient starts persist through 2027, CRISPR's cash burn outlives CASGEVY commercialization.

What makes CRISPR Therapeutics unique

  • CASGEVY is the first approved CRISPR/Cas9 therapy, with 39-country authorization by August 2026.
  • CRISPR built a deep in vivo pipeline: CTX310, CTX340, CTX321, and CTX460.
  • Vertex partnership monetizes CASGEVY globally, while CRISPR retains exposure to upside royalties.
